应用

神经氨酸酶是一种重要的去糖基化酶,能够通过水解 α (2→6)、α (2→3)、α (2→8) 和 α (2→9) 键(按给定的顺序亲和力)裂解所有非还原的不分枝 N-乙酰神经氨酸和 N-羟乙酰神经氨酸残基。使用高浓度的酶和延长孵育时间也可裂解分支的唾液酸。脱唾液酸糖蛋白可通过用各种外切糖苷酶处理进一步表征,导致部分或完全 O-脱糖基化。SDS-PAGE 和 MALDI-TOF MS 通常用于纯化、结构分析和测序过程。这些技术还可去除糖蛋白的异质性和电荷。
