白蛋白基因定位于人染色体 4q13.3。在肝脏中,它合成时是具有 609 个氨基酸的白蛋白前体。通过信号肽的切割产生具有 585 个残基的成熟白蛋白。白蛋白是血清中主要的循环蛋白。它包含三个结构域(I、II 和 III),每个结构域又细分为两个亚结构域 A 和 B。与白蛋白相关配体结合位点总共有 6 个。结构域 IIA 和 IIIA 带有小的杂环或芳香族化合物结合位点。长链脂肪酸结合位点位于 IB 和 IIIB 结构域中。

应用

糖化人白蛋白已用于:
  • 作为用于视网膜小胶质细胞活化的无血清培养基的成分
  • 刺激永生化的近端小管上皮细胞(HK-2)产生纤连蛋白
  • 作为人视网膜微血管内皮细胞(HRMEC)培养物的培养基补充剂,以测试其对序列相似家族 18 成员 B(FAM18B)表达的影响


天然存在的糖化白蛋白是糖尿病患者血糖控制的敏感指标。

生化/生理作用

白蛋白对于生理过程至关重要,并且是脂质、金属离子和氨基酸的主要转运蛋白。它具有抗氧化功能,并能与药物结合。白蛋白基因的突变与常染色体隐性遗传病(称为白蛋白血症)有关,其特征是白蛋白水平较低。白蛋白主要糖基化位点是赖氨酸 525。糖化白蛋白是糖尿病患者血糖的标志物,可用于评估基于 HbA1c 的血糖控制。
