ImmunoHistoMount is a permanent aqueous mounting medium designed for tissue sections and cell smears with peroxidase and alkaline phosphatase chromogens that cannot be dehydrated with organic solvents. This mounting medium preserves fast red, aminoethylycarbazole (AEC), BCIP/NBT, and BCIP/INT chromogens and is compatible with counterstains like hematoxylin and nuclear fast red (NFR). ImmunoHistoMount is also suitable for chromogens like DAB and DAB with nickel and cobalt. It is not compatible with routine H & E staining.

  1. How does the storage temperature relate to shipping conditions?

    The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.   6. At what thickness should the slides be cut for use with ImmunoHistoMount (Product No. I1161)?

    Typically slides are cut at between 5 to 8 microns.

  7. Is ImmunoHistoMount (Product No. I1161) compatible with slides stained with hematoxylin and eosin?

    Slides stained with hematoxylin and eosin should not be coverslipped with ImmunoHistoMount.

  8. What is the step-by-step procedure for using ImmunHistoMount (Product No. I1161)?

    1. Bring the vial to room temperature.  2. Rinse slide to be mounted with Distilled or Deionized water, then touch the edges of slide on a paper towel to remove excess water. Place slides on a flat surface. 3. Turn the vial upside down and open the dropper to remove any air bubbles. 4. Apply 3-4 drops of mounting medium directly on top of the specimen and spread out evenly by tilting the slide back and forth to spread evenly, or use a 0.2 ml plastic pipette tip to evenly distribute the mounting media, making sure the tissue is not touched. Excess medium can be removed by touching the edges of the slide against a paper towel. 5. Let stand at room temperature for about 1 to 2 hours. Alternatively, the slides can be heated at 37-40°C for 40-60 minutes. After setting, the slides are ready for visualization under a microscope.

  9. What are the suggested way to view slides that have been mounted with ImmunoHistoMount (Product No. I1161) under an oil immersion lens?

    There are two ways to look at slides under an oil immersion lens: 1. For oil immersion lens, a drop of oil can be placed directly on dried ImmunoHistoMount mounting medium for visualization. After use, the oil can be removed by wiping with Kimwipes. 2. Apply 2-3 drops of any organic mounting medium on top of dried ImmunoHistoMount mounting medium; which has protected the specimen, coverslip slide with coverslip; the slide is ready for visualization.  

  10. ImmunoHistoMount (Product No. I1161) is stated to be an aqueous mounting media.  Will slides quickly fade?

    ImmunoHistoMount is considered a permanent aqueous mounting media.  Although all slides will eventually fade, slides coverslipped with ImmunoHistoMount should maintain staining for a relatively long period of time.

  11. Can slides to be coverslipped with ImmunoHistoMount (Product No. I1161) be processed through organic solvents?

    Slides to be coverslipped with ImmunHistoMount cannot be dehydrated through organic solvents.
