一般描述

盐酸胍(GdnHCl)是一种广泛使用的蛋白和酶变性剂。分子量小,且天然具有吸湿性。

应用

盐酸胍(GdnHCl)已用作Schlemm管(SC)细胞孵育磷酸盐缓冲液的组分。还用作消化缓冲液组分,消化后的人HeLa229细胞可凝胶化并膨胀(进行膨胀显微镜观察)。盐酸胍还用于分离RNA。
强效离液剂,用于蛋白质的变性以及后续复性。该强效变性剂可溶解不溶性或变性的蛋白质,如包涵体。可用作使蛋白质或酶复性为活性形式的第一步骤。可能还需要脲和二硫苏糖醇 (DTT)。

生化/生理作用

盐酸胍(GdnHCl)可作为酶失活剂,导致酶的构象改变和活性丧失。它参与杂色鲍鱼体内的碱性磷酸酶(ALPase)失活。盐酸胍可在体内阻断热休克蛋白104(Hsp104)腺苷三磷酸酶(ATPase)的活性。还可令木瓜蛋白酶和氨基酰化酶等酶失活。较高浓度的盐酸胍可导致单纯疱疹病毒1型(HSV-1)等病毒灭活。
