除垢剂是一类通过溶解疏水分子降低液液或液固之间的表面张力的表面活性剂或化合物。这些水溶性表面活性剂由疏水性部分组成,通常是一个长的烷基链,与亲水性或水溶性增强的官能团相连。在生物化学、细胞生物学和分子生物学研究中常用于细胞裂解、膜蛋白和脂质纯化、蛋白结晶,以及在免疫印迹实验中减少背景染色。

离子去污剂

离子除垢剂通常含有阴离子或阳离子头部基团及一个净电荷。其疏水尾部不是十二烷基硫酸钠(SDS)和十六烷基三甲基溴化铵(CTAB)等可见的直链烃,就是胆汁酸盐等包含的刚性甾体基团。离子除垢剂溶解膜蛋白非常有效,但基本都会导致一定程度的变性。胆汁酸盐是一种阴离子除垢剂,其主链由刚性甾体基团组成,例如胆酸钠盐和脱氧胆酸钠盐等。这类分子的平面结构使其具有一个极性面和一个非极性面,因此,其CMC较高,胶束较小,易于通过透析去除。

非离子去污剂

非离子除垢剂含不带电的亲水头部基团,该基团由Non-ionic detergents 等除垢剂中的聚氧乙烯部分或辛基葡糖苷和十二烷基麦芽糖苷等中的糖苷基团组成。非离子除垢剂会破坏脂质-脂质和脂质-蛋白互作,但不会破坏蛋白-蛋白的互作,所以是公认的非变性除垢剂,因此广泛应用于分离生物活性形式的膜蛋白。另外,与离子除垢剂不同,非离子除垢剂的胶束尺寸受盐的影响很小。

两性离子去污剂

两性离子除垢剂 兼具离子和非离子除垢剂的特性。与非离子除垢剂一样,两性离子除垢剂不具有净电荷,缺乏导电性和电泳迁移率,且不与离子交换树脂结合。因此是离子交换层析、电泳和等电聚焦中常用的非离子除垢剂的替代物。然而,像离子洗涤剂一样,它们能有效地打破蛋白质与蛋白质之间相互作用。另外,甾体类两性离子除垢剂(如CHAPS)相比直链两性离子除垢剂(如十二烷基二甲基二氧化胺)变性能力通常更弱。
