General description
萤火虫荧光素酶是基因表达中最常用的报告基因之一。这是一种极度灵敏、响应快速且易于使用的报告基因。荧光素酶催化的化学发光反应是测量基因表达最敏感的分析工具之一。与传统方法相比,荧光素酶作用物含有辅酶 A,可增加和维持发光。这可避免向自动光度计进样底物,并允许通过摄影胶片或闪烁计数法进行分析。裂解液含有多粘菌素 B,用于进一步增强信号并消除溶菌酶处理和细胞冻融。细胞裂解液兼容β-半乳糖苷酶测定。由荧光素酶报告基因编码的酶在ATP、氧和Mg2+环境下催化D-萤光素的氧化。定量荧光产物以测定其活性。
