应用

All FSL constructs disperse in biocompatible media and spontaneously and stably incorporate into cell membranes. Cells modified with FSL constructs are known as kodecytes and usually maintain their normal vitality and functionality. FSL-fluorescein has been specifically designed to create fluorescein-labeled live cells (kodecytes) but can also be used to modify other hydrophobic surfaces including fixed cells, virions, and solid phase surfaces. Fluorescein-labeled kodecytes can be visualized at the approximate absorbance wavelength of 488 nm and emission at 518 nm.

FSL-fluorescein is a KODE technology construct designed to label hydrophobic surfaces including living cells with biotin. All FSL constructs consist of three essential designable features:
  • functional component (F)
  • spacer (S)
  • diacyl lipid (L)
FSL-fluorescein is comprised of a fluorescein residue representing F, conjugated via a spacer (SA2) to an an activated adipate derivative of dioleoylphosphatidylethanolamine (L1).
Packaged as 1 mg FSL-Fluorescein with 0.42 mg sodium bicarbonate to improve solubility.

    We recommend reconstituting 1 mg of product F1058, FSL-Fluorescein with 0.5ml of saline or PBS for a concentration of 2 mg/ml.

  6. May I use serum with the product F1058, FSL-Fluorescein?

    We do not recommend diluting the product F1058, FSL-Fluorescein, in lipid-containing buffers. The FSL will associate with the lipids and reduce the efficiency of insertion into cell membranes.

  7. Will the product F1058, FSL-Fluorescein label dead cells?

    The product F1058, FSL-Fluorescein, is designed to insert into the membranes of live cells. But it can also be used to label fixed cells, enveloped virions, and solid phase surfaces.

  8. What concentration of F1058, FSL-Fluorescein do I use to label cells?

    For cell labeling we recommend using a concentration of 10-100ug/ml of F1058, FSL-Fluorescein diluted in PBS. Label cells with FSL-Fluorescein in PBS at a 1:1 volume to volume ratio. The exact concentration may need to be optimized for the cells and the application. Keep incubations in the dark.

  9. How many cells can I label with product F1058, FSL-Fluorescein?

    The exact number of cells may vary depending on cell type and cell condition. The recommended concentration to use for labeling is 10-100ug/ml (~7.4 mM-74mM), which is mixed in a 1:1 volume with cells.
