Biochem/physiol Actions

Preparation Note
该产品是从小鼠Engelbreth-Holm-Swarm肉瘤中提取的；并经过氯仿透析。使用前，将凝胶置于2-8℃下解冻过夜，并使用预冷至2-8℃的板和移液器将凝胶分配至多孔板。也可使用2-8℃的Dulbecco改良Eagle培养基将凝胶稀释最多两倍，应先稀释、再分配至多孔板。 该产品在20°C下放置5分钟即可成胶。对于长时间操作，应在低于10°C的温度下进行。细胞可接种于0.5 mm厚的凝胶层上或在1mm厚的凝胶层内培养。 如果在凝胶层内培养细胞，请在接种前将细胞加至凝胶中，建议密度为3-4 x 104 个细胞/mL。 可使用溶剂为PBS，不含钙、镁或EDTA且浓度为0.6-2.4单位/mL的蛋白酶，将细胞从凝胶中分离出来。
Other Notes
ECM制剂的蛋白质浓度为8-12 mg/mL（DMEM溶液），主要成分为层粘连蛋白，还含有IV型胶原蛋白、蛋白多糖硫酸乙酰肝素、巢蛋白等其他次要成分。向ECM凝胶中添加IV型胶原蛋白可增加其聚合度。
