应用

ECM凝胶可用于上皮细胞、内皮细胞、肌肉细胞、神经细胞和肿瘤细胞。

生化/生理作用

当达到20-40°C时,ECM基质胶将发生热活化聚合,形成重组基底膜。 当PC12细胞在该ECM胶薄层上生长时,将在2天内形成神经突。

用于生产本品的每个小鼠种群都定期筛查病原体。经检测,LDEV呈阴性。

组分

ECM制剂的蛋白质浓度为8-12 mg/mL(DMEM溶液),主要成分为层粘连蛋白,还含有IV型胶原蛋白、蛋白多糖硫酸乙酰肝素、巢蛋白等其他次要成分。向ECM凝胶中添加IV型胶原蛋白可增加其聚合度。

本品含有50 mg/L硫酸庆大霉素。

注意

如需长期储存,请将本品置于-20°C储存。ECM凝胶可在2-8℃下储存最长72小时。

制备说明

该产品是从小鼠Engelbreth-Holm-Swarm肉瘤中提取的;并经过氯仿透析。使用前,将凝胶置于2-8℃下解冻过夜,并使用预冷至2-8℃的板和移液器将凝胶分配至多孔板。也可使用2-8℃的Dulbecco改良Eagle培养基将凝胶稀释最多两倍,应先稀释、再分配至多孔板。 该产品在20°C下放置5分钟即可成胶。对于长时间操作,应在低于10°C的温度下进行。细胞可接种于0.5 mm厚的凝胶层上或在1mm厚的凝胶层内培养。 如果在凝胶层内培养细胞,请在接种前将细胞加至凝胶中,建议密度为3-4 x 104 个细胞/mL。 可使用溶剂为PBS,不含钙、镁或EDTA且浓度为0.6-2.4单位/mL的蛋白酶,将细胞从凝胶中分离出来。

    ECM gel can be diluted twofold with cold Dulbecco's Modified Eagle's Medium (DME).  This means 1 part ECM gel and 1 part DME.

    Product No. E1270, ECM gel is the equivalent of Matrigel from BD only produced by Sigma-Aldrich.

  5. Are Products E1270 and E6909, ECM Gel from Engelbreth-Holm-Swarm murine sarcoma, growth factor reduced like Matrigel (from BD)?

    Product No. E1270 is not growth factor reduced. Product No. E6909 is ECM gel, growth factor reduced.

  6. Do Products E1270 and E6909, ECM Gel from Engelbreth-Holm-Swarm murine sarcoma, contain phenol red?

    Product E1270 ECM gel does contain phenol-red from the Dulbecco's modified Eagle's medium component of the product.  Product No. E6909, ECM gel, growth factor reduced, is phenol red free.

  7. What is the calcium concentration in Products E1270, ECM Gel from Engelbreth-Holm-Swarm murine sarcoma?

    The approximate concentration of CaCl2 in the ECM solution is 0.265 mg/mL.

  8. How can I retrieve my cells from Product E1270,  ECM Gel from Engelbreth-Holm-Swarm murine sarcoma?

    Dispase (Product No. D4818) is a Bacillus-derived neutral protease that is recommended for recovering cells cultured on ECM (Product No. E1270) or Matrigel. Dispase will yield a single cell suspension far more effectively than trypsin, collagenase, or other proteolytic enzymes. Dispase is gentle and will not damage cells harvested for sub-cultivation or other study.
