一般描述

二硫苏糖醇 (DTT) 是维持蛋白质中 SH 基团还原态的优异试剂。它可引发硫醇-二硫键交换反应,完全还原生物分子中的分子内或分子间二硫键。DTT 专门靶向 N,N′-(丙烯酰)胱胺交联剂的二硫键,分解聚丙烯酰胺凝胶和十二烷基聚丙烯酰胺钠凝胶电泳 (SDS-PAGE) 基质。因此,其与定量还原二硫化物的缓冲液联用时非常有效。DTT 的刺激性和毒性低于2-巯基乙醇,浓度也达到β-巯基乙醇的7倍。

应用

DL-二硫苏糖醇用于蛋白质样品中,以在将样品上样到凝胶上进行蛋白质印迹或 SDS-PAGE 之前还原二硫键它已用于 mRNA 原位杂交过程。
