一般描述
免疫球蛋白G（IgG）属于免疫球蛋白家族，是一种广泛表达的血清抗体。这是一种糖蛋白，主要帮助免疫防御系统。大约70％的总免疫球蛋白由IgG组成。IgG是一种单体免疫球蛋白，由四条多肽链组成。它由两条50 kDa重链（γ）和两条25 kDa轻链组成。这些链通过二硫键相互连接。每个分子有两个抗原结合位点。IgG是最丰富的免疫球蛋白。它在血液和组织液中的分布大致相等，占人血清免疫球蛋白的75%。有4个子类：IgG1（66%），IgG2（23%），IgG3（7%）和IgG4（4%）。
特异性
该抗体与人IgG的重链和大多数人免疫球蛋白共有的轻链反应。
免疫原
由纯化人IgG制备。
应用
山羊抗人IgG抗体，HRP偶联物已用于：
- 在酶联免疫吸附测定（ELISA）中检测结合的免疫球蛋白G（IgG）
- 在免疫组化或细胞化学中，稀释度为1：500-1：5,000
- 用于稀释度为1：5,000-1：100,000的显色底物和稀释度为的1：10,000-1：200,000的ECL底物的蛋白质印迹。
- 在EIA中，稀释度为1：5,000-1：100,000
生化/生理作用
免疫球蛋白G（IgG）参与II型和III型超敏反应。它启动补体系统的经典途径，也中和病毒颗粒和毒素。母体IgG可通过人胎盘传递给胎儿，为新生儿抵抗感染提供免疫防御。它促进抗体依赖性细胞介导的细胞毒性（ADCC）。IgG被木瓜蛋白酶消化产生抗原结合片段（Fab）。IgG的胃蛋白酶消化导致可结晶片段（Fc）的形成。严重急性呼吸综合征冠状病毒（SARS-CoV）-2感染患者的IgG水平可能有助于评估2019冠状病毒病（COVID-19）的严重程度和预后。
外形
使用与琼脂糖珠偶联的抗原通过免疫亲和层析纯化。冻干。缓冲液为0.01 M磷酸钠，0.25 M NaCl，pH值7.6和15 mg/ml BSA。不含防腐剂。
储存及稳定性
在接收之日起，将冻干产品在产物2°-8°C下保存长达12个月。复溶后，产品作为未稀释的液体在2°-8°C下稳定6周。用2 mL蒸馏水复溶小瓶。要延长复溶后溶液的保存时间，请添加等体积的甘油，使甘油的最终浓度达到 50％，然后以未稀释的等分试样置于 -20°C 下可保存长达 12 个月。请注意，添加甘油后，抗体（和缓冲盐）的浓度将降低至原浓度的一半，并且必须相应地调整最终的工作稀释度。应避免反复冻融循环。
