特异性

Clone 4D12 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Human leukocyte antigen DRB1 (HLA-DRB1). It targets an epitope within 15 amino acids from the N-terminal extracellular domain of the protein.

应用

Quality Control Testing

Evaluated by Western Blotting in Ramos cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HLA-DRB1 in Ramos cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected HLA-DRB1 in lysates from human PBMCs and human tonsil tissue.

Affinity Binding Assay:: The binding affinity of this antibody to HLA-DRB1 peptide was at least one thousand-fold (1,000X) higher than that obtained with the non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HLA-DRB1 in Raji and Ramos cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected HLA-DRB1 in human PBMC.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected HLA-DRB1 in human tonsil tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

HLA class II histocompatibility antigen, DRB1 beta chain (UniProt: P01911; also known as Human leukocyte antigen DRB1) is encoded by the HLA-DRB1 gene (Gene ID: 3123) in human. HLA-DRB1 is a protein involved in the immune system′s recognition and presentation of foreign antigens to T cells. It belongs to the major histocompatibility complex (MHC) class II proteins. It consists of an alpha chain and a beta chain. The beta chain, encoded by the HLA-DRB1 gene, contains extracellular domains responsible for antigen binding and presentation to T cells. The primary function of HLA-DRB1 is to present antigenic peptides to CD4+ T cells, facilitating immune responses against pathogens. It plays a crucial role in adaptive immunity by presenting antigens derived from external sources, such as viruses or bacteria, to T lymphocytes. HLA-DRB1 is expressed on antigen-presenting cells (APCs) like dendritic cells, macrophages, and B cells. Its expression is regulated by various factors, including cytokines and immune stimulation. In different tissues, HLA-DRB1 expression can vary, indicating tissue-specific roles in immune responses. It forms complexes with other MHC class II molecules, chaperones, and accessory proteins in the endosomal-lysosomal pathway to facilitate antigen processing and presentation. HLA-DRB1 is associated with several autoimmune diseases, including rheumatoid arthritis and certain forms of juvenile idiopathic arthritis. Specific HLA-DRB1 alleles confer susceptibility or protection to these diseases and contribute to the pathogenesis by influencing antigen presentation and immune responses. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

外形

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
