一般描述

(+)-JQ1 是 BET 溴结构域蛋白（包括 BRD2、BRD3、BRD4 和 BRDT）的高亲和力、强效和选择性抑制剂。(+)-JQ1（也称为 SGCBD01）是 (+/-)-JQ1 的活性对映异构体，可抑制 Brd4（含溴结构域 4），通过两个串联的溴结构域（BD1 和 BD2）与染色质形成复合物，这两个溴结构域与组蛋白中的乙酰化赖氨酸残基结合，Brd4 与乙酰化染色质结合，从而可调节延伸因子 b 和其他转录因子对特定启动子区域的募集。已知睾丸核蛋白 (NUT) 基因可与 Brd4 形成融合，从而产生一种有效的致癌基因，导致罕见但高度致死的肿瘤，称为 NUT 中线癌 (NMC)。(+)-JQ1 可抑制 Brd4 与 TNFa 和 E-选择素启动子元件的募集和结合，并在使用 GFP-Brd4 的 FRAP（光漂白后荧光恢复）试验中加速恢复时间。因此 (+)-JQ1 是研究 Brd4 在转录起始中作用的有用工具。
