一般描述

应用
线粒体膜电位试剂盒已用于检测 PC-12 细胞的线粒体膜电位 (MMP) 。
适用性
本试剂盒适于哺乳动物细胞中线粒体跨膜电势差(MMP)的流式细胞检测，并可用于筛选凋亡抑制剂及激活剂等。
原理
该试剂盒使用 JC-10（一种优于 JC-1 的替代品），用于测定细胞中 MMP 的丢失。尽管 JC-1 在许多实验室中广泛使用，但当使用较高浓度时，其水溶性差通常会导致水性缓冲液中出现沉淀。在较高浓度下，JC-10 的水溶性大于 JC-1。与 JC-1 相似，JC-10 是一种阳离子亲脂性染料，浓缩后形成可逆的红色荧光 JC-10 聚集体（λex = 540/ζ em = 590 nm）的细胞线粒体中具有极化的线粒体膜。在凋亡细胞中，MMP 崩解导致 JC-10 不能保留在线粒体中，并使染料恢复其单体、绿色荧光形式（λex = 490/ζ em = 525 nm）。本试剂盒可用于监测细胞凋亡，筛选凋亡抑制剂和激活剂。
