一般描述

适用性

原理

该试剂盒利用 JC-10,一种优于JC-1的替代品,用于测定细胞中MMP的损失。尽管 JC-1在许多实验室中被广泛使用,但是当其在较高浓度下使用时,其水溶性差通常会导致水性缓冲液沉淀。在较高浓度下,JC-10 表现出比 JC-1 更高的水溶性。与JC-1 类似,JC-10是一种阳离子亲脂性染料,其浓缩并在具有极化线粒体膜的细胞线粒体中形成可逆的红色荧光 JC-10 聚集体(λex = 540/λem = 590 nm)。在凋亡细胞中,MMP 崩溃导致 JC-10 无法保留在线粒体中,并使染料返回到单体绿色荧光形式(λex = 490/λem = 525 nm)。该试剂盒可用于监测细胞凋亡和筛选凋亡抑制剂和激活剂。
