一般描述
异柠檬酸脱氢酶 (IDH) 催化异柠檬酸转化为 α-酮戊二酸。在真核生物中,IDH 有三种同工酶,线粒体 IDH2 和 IDH3 以及细胞质/过氧化物酶体 IDH1。所有三个 IDH 家族成员的酶活性均需要存在二价阳离子(Mg 2 + 或 Mn 2 + )和电子接受辅助因子 NADP + (IDH1 和 IDH2)或 NAD + (IDH3)。在某些癌症如胶质母细胞瘤、急性髓性白血病和结肠癌中发现 IDH1 和 IDH2 突变导致新形成层酶活性。该新活性显示底物特异性发生变化,导致 α-酮戊二酸转化为 2-羟基戊二酸。IDH 家族成员的突变也与 Ollier 病和 Maffucci 综合征有关。

原理
异柠檬酸脱氢酶活性测定试剂盒为测定多种样品中 NADP + 依赖性、NAD + 依赖性或 NADP + 和 NAD + 依赖性 IDH 活性提供了一种简单直接的方法。在酶反应中使用异柠檬酸盐作为底物测定 IDH 活性,得到比色产物 (450 nm) 与存在的酶活性成正比。一个单位的 IDH 是在 37°C、pH 8.0 时每分钟生成 1.0 μmol NADH 或 NADP 的酶量。
