溶酶体分离试剂盒可通过差速离心后进行密度梯度离心和/或钙沉淀，将溶酶体从动物组织和培养细胞中有效分离出来。是否存在溶酶体可通过使用酸性磷酸酶测定试剂盒（货号 CS0740）测定。Sigma提供合适的标记物检测试剂盒，可检测与其他细胞器的分离情况。
