一般描述
谷胱甘肽还原酶(GLR1)在线粒体和细胞质中存在异构体。它和硫氧还蛋白还原酶有相同的序列,结构同源,是含有黄素的氧化还原酶。它的活性位点是由氧化还原活性二硫组成,催化活性需要NADPH。这种酶广泛存在于植物,细菌,酵母,小鼠和人类。
应用
来自烘焙′s 酵母(Saccharomyces cerevisiae) 的谷胱甘肽还原酶(GR)用于
- 使用谷胱甘肽还原酶-5,5′-二硫基双(2-硝基苯甲酸)-基酶回收实分析定量心肌组织中的谷胱甘肽含量。
- 使用略微改良的微谷胱甘肽分析,定量初情期母猪卵母细胞中的还原谷胱甘肽(GSH)
- 制备总GSSG(谷胱甘肽二硫化物)+GSH检测,然后在大鼠晶状体内所有GSSG被还原成GSH。

生化/生理作用
谷胱甘肽还原酶 IGR) 是抗氧化系统中关键的黄素酶。谷胱甘肽超氧化物酶使用还原谷胱甘肽(GSH),去除过氧化氢毒性,在这一过程中GSH会被转化成氧化谷胱甘肽(GSSG)。然后GSSG被谷胱甘肽还原酶(GR)回收变成GSH,这一过程中NADPH变成NADP+。 重生的GSH能够解更多的过氧化氢的毒。该酶使用FAD作为辅因子。GR和谷胱甘肽过氧化物酶可能在精子中作为抗氧化酶,抑制脂质过氧化。 谷胱甘肽还原酶和其他一些蛋白质有共同的结构模体,包括天冬氨酸蛋白酶、柠檬酸合成酶、EF手性分子、血红蛋白、脂质体和α/β水解酶。 GR被褪黑素激活,被报道不可逆地被一些氧自由基产生系统抑制。
谷胱甘肽(γ-谷氨酰半胱氨酸甘氨酸)是一种广泛存在的三肽硫醇,在细胞氧化压力抵抗机制中起到关键作用。谷胱甘肽还原酶(GLR1)负责还原谷胱甘肽二硫化物(GSSG),形成还原谷胱甘肽。
