一般描述

纤溶酶原激活物抑制剂1 (PAI-1)或丝氨酸蛋白酶抑制剂E1 (SERPINE1)基因映射到人类染色体7q22.1。PAI-1在肝细胞、内皮细胞、平滑肌细胞、巨核细胞和脂肪细胞中产生。

应用

人纤溶酶原激活物抑制剂1 (PAI-1)已被用作:
  • 放射免疫沉淀法(RIPA)中的脑源性神经营养因子(proBDNF)的抑制剂,用于外周血和淋巴细胞
  • 阻断感染白色念珠菌菌丝
  • 小鼠的纤溶酶活性,检测其对神经元神经突生长的影响

生化/生理作用

纤溶酶原激活物抑制剂1 (PAI -1)参与介导细胞衰老。它在损伤期间与其他组织修复转录组一起上调。PAI-1水平升高被认为是与心肌梗死(MI)相关的风险之一。
纤溶酶原激活物抑制剂1(PAI-1)蛋白属于丝氨酸蛋白酶抑制剂的Serpin超家族。它是组织纤溶酶原激活物(tPA)和尿激酶(uPA)的主要抑制剂,并也是纤溶酶原的激活剂,因此可抑制纤维蛋白溶解过程。 PAI可参与到细胞外基质的重塑,并涉及血管生成、趋化性、排卵和胚胎生成等过程。 PAI-1在各种疾病状态中的水平都有所上升,如多种癌症形式、肥胖症和代谢综合征。
