革兰氏番红溶液，又称番红T或碱性红2，是一种即用的染色溶液，包含10%的乙醇番红。番红是一种碱性、阳离子、偶氮染料，含有一个平面的、二氨基吩嗪发色团，苯基取代基连接到一个环氮上。番红的三甲基阳离子形式是亲脂性的，而二甲基形式是弱亲水性的。它最常用于细菌学中的革兰氏染色复染，以区分革兰氏阳性菌和革兰氏阴性菌。

应用

革兰氏番红溶液已用于以下应用：
  • 作为无毒结晶紫替代品，用于生物膜形成的定量
  • 开发透明质酸可视化方法
  • 研究如何通过2-氨基噻唑改善β-内酰胺对生物膜内嵌金黄色葡萄球菌的敏感性。

生化/生理作用

革兰氏染色可区分革兰氏阳性和革兰氏阴性细菌。细菌细胞壁的胞壁质结构是颜色亲和力的基础。首先用结晶紫给细菌染色，然后用碘溶液染色，形成染料-碘复合物。这种复合物在脱色后保留在革兰氏阳性菌细胞壁的多层粘肽结构中，形成蓝紫色。 革兰氏阴性细菌的细胞壁由单层粘肽结构组成，不会保留染料-碘复合物，可吸收番红复染液，从而呈现粉红色。
