一般描述

在细胞凋亡的早期阶段,细胞表面会发生变化。这些质膜的改变之一是磷脂酰丝氨酸(PS)从质膜内侧移位到外层,从而将PS暴露于细胞表面。巨噬细胞可特异性识别在凋亡发展过程中暴露于淋巴细胞表面的PS。凋亡细胞和身体的识别和吞噬作用可保护生物免于暴露于导致炎症的细胞化合物中,这些化合物通常会导致细胞坏死。膜联蛋白V是一种35.8kDa,Ca2+ -依赖性磷脂结合蛋白,其对PS具有高亲和力,这使得标记的膜联蛋白成为优异的检测剂。

检测:膜联蛋白-V-FLUOS可以在FACS分析和免疫化学中直接进行检测,无需二级检测系统。

特异性

膜联蛋白-V-FLUOS以Ca2+依赖形式与带负电荷的磷脂表面进行结合,并且对磷脂酰丝氨酸表现出高特异性。因此,它可对凋亡和坏死细胞进行染色。碘化丙锭仅可对渗漏的坏死细胞的DNA进行染色,并可区分凋亡细胞和坏死细胞。

应用

经标记的膜联蛋白-V具有对磷脂酰丝氨酸(PS)的高亲和力,可作为对暴露于凋亡细胞外层的PS的敏感探针。由于坏死细胞膜完整性的丧失,也会导致PS的暴露,因此碘化丙啶被用作DNA染色以区分坏死细胞和膜联蛋白-V标记的细胞簇。其他二级标记也可能适用,例如用藻红蛋白或TRITC标记的单克隆抗体进行膜表面染色以进一步进行细胞表征。

制备说明

工作溶液:在1ml孵育缓冲液中预稀释20μl膜联蛋白-V-FLUOS标记试剂,并加入20μl碘化丙啶溶液。
注:1ml足够用于10个样品。
储存条件(工作溶液):稀释的染色溶液应该包装新鲜制备。
