原理

预先用 BrdU 标记的样品用乙醇固定,然后用 BrdU 单克隆抗体孵育,该抗体含有优化的核酸酶混合物。这些核酸酶产生单链 DNA 片段,允许抗体与 BrdU 结合。接着,加入碱性磷酸酶 (AP) 标记的小鼠免疫球蛋白抗体,然后与抗 BrdU 抗体结合。然后将样品与 AP 底物和代谢形成有色反应产物的 NBT/BCIP 一起孵育。使用相差显微镜评价样品。

制备说明
