一般描述

DNase I是一种可降解DNA的双链特异性内切核酸酶。牛胰腺脱氧核糖核酸酶I(DNase I)是DNA小沟相互作用的核酸酶,特异性相对较低。该蛋白质由两个中心β折叠构成,而每个折叠由六个β-链所组成。该结构被大量的环以及α-螺旋区域包围。该酶与核酸外切酶III具有结构相似性。DNase I是研究得最透彻的哺乳动物来源的内切酶之一。

应用

DNase I 与胶原酶一起已被用于组织解离。

生化/生理作用

在组织解离过程中,部分细胞被裂解并导致DNA的释放。单分子DNA可能会导致细胞结团。向解离缓冲液中加入DNase I会导致该细胞外DNA的降解,因而避免了因意外结团造成的细胞损失。

制备说明

激活剂:二价金属离子 (Ca2+, Mg2+)

重悬

用无菌的双蒸水对冻干酶进行重悬(10 mg/ml);用PBS(磷酸盐缓冲液)、HBSS(Hank′s 平衡盐溶液)或培养基进行进一步的稀释。
