MEKK5 (MAP3K5) is a key kinase for our innate immunity and for mediating cellular signaling cascades evoked by changes in the external cellular environment. As such, MEKK5 is critical for cell fate during development, apoptosis, and cell survival as well as in modulating the host defense responses from a wide range of pathogens. MEKK5 acts as an upstream activator of the MKK/JNK signaling pathways and the p38 MAPK signaling pathways that ultimately results in activating multiple kinases such as MAP2K3/Mkk3 or MKK6/MMK7. Activation of MMEKK5 is via phosphorylation of Thr-845 within the activation loop via the recently discovered kinase MAP3K6 which forms a complex with MEKK5 and also stabilizes the protein. Expression is widespread and localized in the cytoplasmic in the perinuclear endoplasmic reticulum region.
