蛋白质印迹:
在新生大鼠或小鼠细胞提取物中的还原性凝胶中200-220 kDa。请注意，巢蛋白最初（错误地）被报告通过（SDS-PAGE）会迁移至大约440 kDa。
对于免疫印迹实验，可通过在4倍体积的125 mM Tris，pH 6.8、10％ 2-巯基乙醇、10％甘油和4.6％ SDS中煮沸而制备样品。推荐使用5% PAGE胶。用牛奶或BSA进行膜封闭。不要用含有牛奶的缓冲液进行一抗稀释。对样品进行冻融或未将样品保存在-85°C，有时会使巢蛋白断裂成多个片段。

免疫组化：
使用了之前一个批次的1:20-1:200稀释液。对于光学显微镜，可用pH 7.4的4％多聚甲醛灌注组织，而对于电子显微镜可用pH 10.0的4％多聚甲醛或用含0.1％戊二醛的pH 7.4的4％多聚甲醛灌注组织。适用于冷冻或石蜡包含切片。

免疫细胞化学：
使用了之前一个批次的1:20-1:200稀释液。用pH 9.5的50 mM硼酸钠缓冲的4％多聚甲醛进行细胞固定。

终端用户需要对最佳的工作稀释比和实验方法进行确定。

General description

巢蛋白，一种在发育过程中以及在肌腱和神经肌肉接头中表达的大型中间丝蛋白（VI型）。巢蛋白表达受到限制，通常在E18时消失。巢蛋白被认为是一种合理的神经元标志物。然而，近期研究发现巢蛋白在其他细胞类型，如内皮细胞中也有所表达（Mokry和Nemcek，1998）。巢蛋白可识别最原始的神经上皮细胞，但也可以识别许多其他胚胎组织，因此它对CNS并不特异。巢蛋白的表达在几乎所有GBM（各种胶质母细胞瘤）和多种黑色素瘤（原发性和转移性）中均可见，但在任何转移性癌症中均未见到。
