整联蛋白αV的分子量：125-135kda
细胞粘附受体的整联蛋白家族由至少16种膜相关的异二聚体组成，由以非共价方式缔合的α和β亚基组成。整联蛋白家族的结构和功能多样性基于各个α和β亚基的配对能力。整联蛋白受体与其各自配体之间这些分子相互作用的关键是对Arg-Gly-Asp（RGD）序列的识别，已知该序列存在于细胞外基质成分包括纤连蛋白、玻连蛋白、胶原蛋白、纤维蛋白原和血管性假血友病因子中（Cheresh，1991）。整联蛋白在血管增殖、粘附和伤口修复中的作用已被充分证明。粘附受体整联蛋白αvβ3似乎在生长的血管上选择性表达，并已被确定为血管生成血管组织的标志物（Brooks，1994）。由于其可参与血管生成，整联蛋白αVβ3是研究最多的整联蛋白受体之一。

免疫原

从人黑素瘤细胞系M21纯化的粘附受体（Cheresh，1987）。

应用

流式细胞术：100μL中每0.5 x 10e6个细胞1-3 μg LM609（Smith，1996）。

免疫沉淀：特异性免疫沉淀整联蛋白αvβ3复合物，当在还原条件下通过SDS-PAGE分离时，产生对应于αv亚基的约130 kDa条带和对应于β3亚基的105 kDa条带（Cheresh，1987）。

免疫荧光：5-10 μg/mL，用于用丙酮固定的经bFGF处理的6 mm鸡胚绒毛尿囊膜冷冻切片的染色（Brooks，1994）。LM609对4%多聚甲醛固定、冷冻的组织和细胞也有效。但对石蜡包埋组织切片的免疫组织化学染色无效。

在10-25 μg/mL浓度下抑制细胞对玻连蛋白包被表面的粘连。

最佳的工作稀释度和方案必须由最终用户确定。

生化/生理作用

与玻连蛋白受体αvβ3复合物（一种RGD定向的粘附受体）反应。已经证明LM609对人黑素瘤细胞系（M21）对玻连蛋白、纤维蛋白原和血管性假血友病因子以及含有肽的合成RGD的阻断粘连（Cheresh，1987）。在鸡胚绒毛尿囊膜中，LM609显示阻断bFGF和TNFα诱导的血管生成，但对已有的血管没有作用（Brooks，1994）。尽管LM609阻断细胞粘附在含有RGD的配体上，但它不直接与RGD结合位点相互作用。相反，LM609似乎是整联蛋白αvβ3的变构抑制剂，其与由αV和β3亚基的翻译后关联性产生的构象表位结合。
