一般描述

免疫印迹是研究蛋白质功能和定位的常用技术。 通常，将蛋白样品在SDS-PAGE上进行电泳，然后转移到硝酸纤维素膜或尼龙膜上，然后在其中用特异性抗体进行探测。 与可重复利用Southern和Northern印迹的核酸技术不同，免疫印迹很难重复使用。

免疫印迹的剥离和重新检测技术具有以下几个优点：

1）保存昂贵或数量有限的样品，

2）使用几种不同的抗体（例如亚型或亚型特异性抗体）分析给定的印迹，

3）重新分析异常结果并使用相同或不同的抗体进行确认，

4）用错误的抗体纠正孵育过程中的错误，

5）通过重复使用相同的印迹节省试剂和时间的成本。

尽管基于抗原和抗体的免疫亲和基质（例如Sepharose™偶联物）已被重复使用多次而不会损害抗原-抗体的反应性，但对极端pH值和离液剂的需求已排除了这些方法在蛋白质印迹中的应用。

MILLIPORE Re-Blot Plus Western Blot强效抗体剥离溶液包含特殊配制的溶液，可快速有效地从蛋白质印迹中去除抗体，而不会显著影响固定的蛋白质。

Re-Blot Plus Western Blot强效抗体剥离液的优点包括：

· 抗体剥离液中不含有刺激性气味的b-巯基乙醇。

· 抗体剥离在室温下进行。无需加热印迹。

· 可在室温下约15分钟内剥离抗体印迹。

· 印迹可在25分钟内重复使用。

应用

MILLIPORE Re-Blot Plus Western Blot强效抗体剥离液可有效去除已用化学发光法或放射性碘或其他同位素显影的蛋白质印迹中的抗体。不建议剥离比色底物（TMB、DAB、4-氯萘酚等），因为不可能有效去除在反应位点沉淀的底物。

在通过比较印迹分析确定剥离不会定量影响给定抗原之前，Re-Blot Plus Western Blot强效抗体剥离溶液应仅用于定性。

本品仅供研究使用；不用于诊断或体内使用。

制备说明

强效的抗体剥离液应在到达后在2-8°C下保存。 产品在接收后可稳定保存3至6个月。 如果抗体剥离液在储存时结晶，则可以在使用前在37°C下缓慢加热将其重新溶解。

注意：为防止试剂降解，在储存时盖紧瓶盖。 避免长时间暴露在空气中。
