共济失调的毛细血管扩张突变激酶（ATM）和共济失调的毛细血管扩张以及Rad3相关激酶（ATR）是调节细胞周期检查点和DNA修复的相关激酶。ATM基因突变会导致常染色体隐性遗传疾病共济失调毛细血管扩张（AT）。已被鉴定的ATM底物包括p53、p95/NBS1、MDM2、Chk2、BRCA1、CtIP、4E-BP1和Chk1。对ATM/ATR底物的基本要求是S/TQ。在-3和-1位的疏水氨基酸和在+1位带负电荷的氨基酸是这些激酶识别底物的阳性决定因素。S/TQ周围带正电荷的残基是底物磷酸化的负决定因素。来自AT患者的细胞的复杂表型表明ATM具有其他细胞底物。在未经辐照的细胞中，ATM以无活性的同二聚体或多聚体形式存在。电离辐射导致的DNA双链断裂通过解离该复合物和在Ser1981处进行ATM自磷酸化而导致ATM激酶的快速活化。

免疫原

KLH偶联的合成肽，对应于人ATM的氨基酸的1974-1988残基（SLAFEEG [pS] QSTTISS）。 免疫序列在小鼠和大鼠中具有11/12的相似度。

应用

使用经过ICC、IF、IP、WB验证的抗磷酸化ATM（Ser1981）抗体，克隆10H11.E12（小鼠单克隆抗体），对磷酸化ATM（Ser1981）（也称为AT突变）进行检测。
免疫沉淀：
从辐照的HeLa细胞中进行免疫沉淀磷酸化的ATM（图A，第3和4泳道）。

免疫细胞化学：
在辐照的人和小鼠成纤维细胞中检测到病灶。由独立实验室进行确定。

该抗体可识别ATM，分子量〜370 kDa。一种非特异性蛋白也会被检测到，分子量〜> 400 kDa。

蛋白G纯化
蛋白G纯化的小鼠IgG，溶于含0.175 M NaCl、0.07％叠氮化钠和30％甘油的0.014 M磷酸盐缓冲液（pH 7.6）中。在-20°C为液体形式。

制备说明

自收到之日起，在 -20°C 条件下可稳定保存 1 年。

处理建议：
收到后，在取下瓶盖之前，将小瓶离心并轻轻混合溶液。分装到微量离心管中，并储存于 -20°C。避免反复冻融循环，以免损坏 IgG 和影响产品性能。注意：冷藏室温度变化至低于-20°C时可能导致含甘油的溶液在储存过程中冻结。

分析说明

对照
经辐照的HeLa细胞裂解物
通过免疫印迹对辐照的HeLa细胞的粗裂解液进行常规评估。

蛋白质印迹分析：
0.5 µg/mL该批次产品可检测到辐照HeLa细胞的粗裂解物中的磷酸化ATM。
