何为抗生素杀灭曲线?

抗生素杀灭曲线是一项剂量响应实验,通过逐渐增大施加在哺乳动物细胞中的备选抗生素剂量,测定特定时间内杀死所有细胞的最低抗生素浓度。这是使用备选抗生素杀死未转染细胞、形成稳定细胞系之前的关键步骤。由于每种哺乳动物细胞系对抗生素的敏感程度不一,因此始终建议针对不同细胞系和抗生素单独进行杀灭曲线实验。

杀灭曲线实验方案

以下实验方案概述了绘制细胞杀灭曲线的步骤。

准备物品:

  • 健康状态的目标细胞系
  • 您的细胞系建议的完全培养基(培养基 + 血清)
  • 您感兴趣的备选抗生素

步骤:

  • 通过胰蛋白酶或温和刮取,收获健康的贴壁细胞。
  • 用完全培养基稀释细胞悬液,接种到 96 孔板各孔中,终体积为 100 ul。处理当天建议的细胞密度约为 50%。
  • 24孔板中抗生素处理前的典型细胞密度:
  • 0.8 – 2.5 X 105各细胞/ml贴壁细胞
  • 2.5 – 4.5 X 105个细胞/ml 悬浮细胞
  • 37 oC孵育过夜,或直至所有细胞均调整良好且健康。

注意:细胞处于活跃分裂状态时,抗生素最有效。 因此,建议采用最优细胞密度,以实现最准确的剂量响应。

  • 用含不同浓度备选抗生素的新鲜培养基置换培养基。参照96孔板形式(图 1)所示,每个浓度维持一式三份。
  • 每48小时更换含抗生素的培养基。
  • 培养细胞7-10天,定期在光学显微镜下观测细胞。根据所用细胞系,培养时间可延长至14天。
  • 第10天,通过MTT测定法或准确的细胞计数器测定各孔细胞活率。
  • 选择完全阻止细胞生长的抗生素浓度。在所有转染实验中,基于该浓度筛选可进一步生长为稳定细胞系的耐受性转染细胞(图2)。

谨记以下要点

  • 维持适当的对照或空白样:
    • 孔中仅含有培养基和抗生素,不含细胞
    • 孔中仅含有培养基和细胞,不含抗生素
  • 确保培养箱处于完美工作状态,避免因培养基蒸发导致抗生素浓度增大,从而导致读数错误
