Quality Control Testing
Evaluated by Western Blotting with recombinant human IL-6R.
Western Blotting: A 1:10,000 dilution of this antibody detected recombinant human IL-6R.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected IL-6R in U266B1 cell lysate.
Flow Cytometry Analysis: 1 µg from a representative lot detected IL-6R in one million human peripheral blood mononuclear cells (PBMC).
Affinity Binding Assay: A representative lot of this antibody bound IL-6R peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IL-6R in U937 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IL-6R in human spleen, human tonsil, human testis cancer, and human liver cancer tissue sections.

Interleukin-6 receptor subunit alpha (IL-6R) (UniProt: P08887; also known as IL-6 receptor subunit alpha, IL-6R subunit alpha, IL-6R-alpha;IL-6RA, IL-6R 1, Membrane glycoprotein 80 (gp80), Soluble interleukin-6 receptor subunit alpha, sIL6R, CD126) is encoded by the IL6R gene (Gene ID: 3570) in human. IL-6R is a component of the IL-6 receptor complex. It composed of an extracellular domain that binds IL-6, a transmembrane domain, and a relatively short cytoplasmic tail that does not have intrinsic signaling capability. Upon Il-6 binding, IL-6R pairs with the glycoprotein 130 (gp130) co-receptor, leading to the activation of various intracellular signaling pathways, including JAK/STAT, MAPK, and PI3K-Akt. These pathways regulate gene expression involved in immune response, inflammation, cell survival, proliferation, and differentiation. IL-6R is expressed in a variety of cell types, including immune cells like T cells, B cells, and macrophages, as well as liver and some endothelial cells. The expression can be regulated by inflammatory stimuli, which enhances the responsiveness of cells to IL-6 during inflammation or infection. IL-6R is primarily localized on the cell surface where it interacts with circulating IL-6. However, a naturally occurring soluble form of IL-6R (sIL-6R) also exists, generated either by alternative splicing or by proteolytic cleavage of the membrane-bound receptor. This soluble receptor can bind IL-6 and activate cells via gp130, even in the absence of membrane-bound IL-6R, a process known as trans-signaling. IL-6R plays a significant role in the pathogenesis of various inflammatory and autoimmune diseases, such as rheumatoid arthritis, Castleman′s disease, and systemic lupus erythematosus. Overactivation of IL-6/IL-6R signaling is linked to chronic inflammation and autoimmunity. Targeting IL-6R with therapeutic antibodies like tocilizumab, which blocks IL-6R, has proven effective in treating these conditions. Additionally, increased levels of sIL-6R are associated with severe asthmatic symptoms and other inflammatory disorders. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Avoid repeated freeze-thaws.
