General description

Glucagon receptor (UniProt: P47871; also known as GL-R) is encoded by the GCGR gene (Gene ID: 2642) in human. The glucagon receptor (GCGR) is a G protein-coupled receptor (GPCR) that consists of seven transmembrane helices. It has an extracellular N-terminus that binds glucagon, a peptide hormone, and an intracellular C-terminus that interacts with G proteins. The receptor undergoes conformational changes upon glucagon binding, activating intracellular signaling pathways. The glucagon receptor is crucial for glucose homeostasis. When glucagon binds to its receptor, it activates adenylate cyclase, leading to an increase in cyclic AMP (cAMP) levels. This cascade results in the stimulation of gluconeogenesis and glycogenolysis in the liver, raising blood glucose levels. Additionally, GCGR plays a role in lipid metabolism and may influence appetite regulation. GCGR is predominantly expressed in the liver, where it exerts its primary metabolic effects. It is also found in lower levels in other tissues, including the pancreas, kidney, and brain. The expression of the glucagon receptor can be influenced by various factors, including hormonal signals and metabolic states. Increased GCGR activity can lead to hyperglycemia and is implicated in type 2 diabetes mellitus. Conversely, decreased receptor function can contribute to hypoglycemia. Additionally, GCGR has been studied as a potential target for therapeutic interventions in obesity and metabolic syndrome. Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the extracellular domain of human Glucagon receptor.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
