Quality Control Testing
Evaluated by Western Blotting in U-251 MG cell lysate.
Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected DKK1 protein in U-251 MG cell lysate.
Tested Applications
Affinity Binding Assay: A representative lot of this antibody bound DKK1 peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected DKK1 in human placenta tissue sections.
Immunocytochemistry Analysis : A 1:100 dilution from a representative lot detected DKK1 in PC-3 cells.

Dickkopf-related protein 1 (UniProt: O94907; also known as Dickkopf-1, Dkk-1, hDkk-1, SK) is encoded by the DKK1 (also known as UNQ492/PRO1008) gene (Gene ID: 22943) in human. Dickkopf-1 (DKK1) is a secreted protein that is a member of the Dickkopf family involved in modulating Wnt signaling pathways. DKK1 contains two cysteine-rich domains connected by a linker region, which are crucial for its function and interaction with other proteins, such as the Wnt co-receptors LRP5 and LRP6. DKK1 functions primarily as an antagonist of the Wnt signaling pathway, a key regulator of cell growth, differentiation, and embryonic development. By binding to LRP5/6, DKK1 prevents the formation of the Wnt-Frizzled-LRP complex, thereby inhibiting the canonical Wnt signaling pathway. This inhibition is critical for regulating processes such as bone formation and growth. DKK1 is expressed in a variety of tissues, with significant roles in both embryonic development and adult tissue homeostasis. Its expression is particularly noted in bone, where it is involved in the regulation of osteoblast and osteoclast activity, impacting bone density and turnover. Overexpression of DKK1 is associated with bone loss and osteoporosis, as it inhibits Wnt signaling which is crucial for bone formation. Conversely, reduced levels of DKK1 are seen in conditions characterized by excessive bone growth. DKK1 is also involved in cancer biology; it can either promote or inhibit tumor growth depending on the cancer type and the context of Wnt signaling activity. In some cancers, such as multiple myeloma, DKK1 expression is linked to poor prognosis due to its role in creating a bone-destructive environment around the tumor. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
