Clone 1D16 is a ZooMAb^® rabbit recombinant monoclonal antibody that specifically detects Nephrin. It targets an epitope within 25 amino acids from the C-terminal, cytoplasmic domain.

Quality Control Testing

Evaluated by Western Blotting in Human kidney tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Nephrin in Human kidney tissue lysate.

Tested Applications:

Immunohistochemistry (Paraffin) Analysis: A 1:1000 dilution from a representative lot detected Nephrin in human kidney tissue sections.

Affinity Binding Assay Analysis: 300 µg/mL of this antibody detected Nephrin in Nephrin 3.2 x 10^-9.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Nephrin in HEK 293 cells.

Nephrin (UniProt: O60500; also known as Renal glomerulus-specific cell adhesion receptor) is encoded by the NPHS1 (also known as NPHN) gene (Gene ID: 4868) in human. Nephrin is a single-pass, type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 23-1055), a transmembrane domain (aa 1056-1076), and a cytoplasmic domain (aa 1077-1241). It is essential for the proper functioning of the renal filtration barrier and regulation of glomerular vascular permeability. It may also anchor the podocyte slit diaphragm to the actin cytoskeleton. It is specifically expressed in podocytes of kidney glomeruli, which along with endothelial cells and the basement membrane forms the glomerular filtration barrier. The glomerular basement membrane functions as a coarse prefilter for large plasma proteins, whereas the slit diaphragm serves as an isoporous ultrafilter with pores

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
