目标描述
Ankyrin-3 (UniProt: Q12955; also known as ANK-3, Ankyrin-G) is encoded by the ANK3 gene (Gene ID: 288) in human. Ankyrins are a family of spectrin-binding scaffolding proteins that couple the spectrin/actin cytoskeletal network to the cytoplasmic domains of integral membrane proteins, which include channels and neuronal cell adhesion molecules. The ankyrin family of proteins consists of Ankyrin-R, Ankyrin-B, and Akyrin-G. Ankyrin-B and ankyrin-G have been detected in a variety of tissue. In contrast, ankyrin-R is detected only in erythrocytes, striated muscle, and some neurons. Ankyrin protein has four functional domains: a membrane-binding domain (MBD), spectrin-binding domain (SBD), death domain, and a C-terminal regulatory domain. Ankyrin-G is a involved in various protein interactions and neuronal functions, most notably the formation and maintenance of the axon initial segment and node of Ranvier of neurons. It links ion channels to the cytoskeleton in these areas and is critical for action potential generation and propagation. Its expression is observed in brain, muscles, and other tissues. Its levels are up-regulated during muscle cell differentiation. It may participate in the maintenance/targeting of ion channels and cell adhesion molecules at the nodes of Ranvier and axonal initial segments. In skeletal muscle it is required for costamere localization of dystrophin (DMD) and β-dystroglycan 1 (β-DAG1). It is reported to directly interact with DMD and β-DAG1, however, this interaction does not interfere with binding between DMD and β-DAG1. Mutations in ANK3 gene have been linked to bipolar disorder and other psychiatric disorders. Five isoforms of Ankyrin-G have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. 外形
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
重悬
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
储存及稳定性
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
