General description

40S ribosomal protein S25 (UniProt: P62851; also known as Small ribosomal subunit protein eS25) is encoded by the RPS25 gene (Gene ID: 6230) in human. RPS25 plays a key role in several forms of unconventional translation including internal ribosome entry site (IRES)-mediated translation and ribosomal shunting. RPS25 is reported to interact with MDM2 and inhibit its E3 ligase activity that results in the reduction of MDM2-mediated p53 ubiquitination leading to p53 stabilization and activation. RPS25, MDM2, and p53 form a ternary complex following ribosomal stress. It is required for efficient RAN translation of C9orf72 and other neurodegenerative disease-associated nucleotide repeats. However, reduction in RPS25 does not alter endogenous C9orf72 protein expression. Reduction in RPS25 levels by siRNA results in reduced levels of poly(GP) without affecting the number of RNA foci or levels of the different C9orf72 alternative transcript variants, including transcripts harboring the GGGGCC repeat. RPS25 also regulates translation initiation of hepatitis C virus and picornaviral IRES RNAs, downstream of 40S subunit recruitment. Application

Quality Control Testing

Evaluated by Western Blotting in Rat Liver tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected RPS25 in Rat Liver tissue lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RPS25 in A549 and NIH3T3 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected RPS25 in human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound recombinant human RPS25 protein with a KD of 4.7 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected RPS25 in HT-29 and U2OS cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

Biochem/physiol Actions

Clone 3G15 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects 40S ribosomal protein S25 (RPS25).

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
