Evaluated by Western Blotting in HepG2 cell lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected RSK-2 in HepG2 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected RSK-2 in A431 and COS-7 cell lysates and 1:1,000 dilution in NIH3T3 and C6 cell lysates.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected RSK-2 in HeLa cells.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected RSK-2 in Human stomach and Human liver tissue sections.

Ribosomal protein S6 kinase alpha-3 (UniProt: P51812; also known as EC:2.7.11.1, S6K-alpha-3, 90 kDa ribosomal protein S6 kinase 3, p90-RSK 3, p90RSK3, Insulin-stimulated protein kinase 1, ISPK-1, MAP kinase-activated protein kinase 1b, MAPK-activated protein kinase 1b, MAPKAP kinase 1b, MAPKAPK-1b, Ribosomal S6 kinase 2, RSK-2, pp90RSK2) is encoded by the RPS6KA3 (also known as ISPK1, MAPKAPK1B, RSK2) gene (Gene ID: 6197) in human. RSK-2 is a serine-threonine protein kinase that is expressed in various tissues with highest levels observed in the skeletal muscle. It acts downstream of ERK2/ERK1 signaling and mediates mitogenic and stress-induced activation of various transcription factors, including CREB1, ETV1/ER81 and NUR77, and regulates translation through RPS6 and EIF4B phosphorylation. It mediates cell proliferation, survival, and differentiation by modulating mTOR signaling and repressing pro-apoptotic function of BAD and death-associated protein kinase 1 (DAPK1). Upon insulin stimulation, it acts indirectly on the transcription regulation of several genes by phosphorylating GSK-3β at serine 9 and inhibiting its activity. RSK-2 is also involved in the mTOR nutrient-sensing pathway by directly phosphorylating TSC2 at serine 1798 that inhibits TSC2 ability to suppress mTOR signaling. It also mediates phosphorylation of RPTOR, that regulates mTORC1 activity and may promote rapamycin-sensitive signaling independently of the PI3K/AKT pathway. RSK-2 is activated by phosphorylation at serine 227 by 3-phosphoinositide dependent protein kinase 1 (PDPK1). 外形
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
