Quality Control Testing
Evaluated by Western Blotting in Human peripheral blood mononuclear cells (PBMC) lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected S100-A8 in lysate from peripheral blood mononuclear cells (PBMC).
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected S100-A8 in THP-1 and HL-60 cell lysates.
Affinity Binding Assay: A representative lot of this antibody bound S100-A8 peptide with a KD of 5.7 x 10-6 in an affinity binding assay.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected S100-A8 in human spleen tissue sections.
Flow Cytometry Analysis: 1 µg from a representative lot detected S100-A8 in one million Human peripheral blood mononuclear cells (PBMC).

Protein S100-A8 (UniProt: P05109; also known as MRP-8, Migration inhibitory factor-related protein 8 Calgranulin-A, Calprotectin L1L subunit, Leukocyte L1 complex light chain, p8, S100 calcium-binding protein A8) is encoded by the S100A8 (also known as MRP8) gene (Gene ID: 6279) in human. Protein S100-A8 is a calcium- and zinc-binding proteins that plays a prominent role in the regulation of inflammatory processes and immune response. It is known to induce neutrophil chemotaxis and adhesion. It is predominantly found as calprotectin and performs a variety of intra- and extracellular functions. Its intracellular functions include facilitating leukocyte arachidonic acid trafficking and metabolism, modulation of the tubulin-dependent cytoskeleton during migration of phagocytes, and activation of the neutrophilic NADPH-oxidase. Protein S100-A8 activates NADPH-oxidase by facilitating the enzyme complex assembly at the cell membrane. It contributes to the enzyme assembly by directly binding to NCF2/P67PHOX. Its extracellular functions include pro-inflammatory, antimicrobial, oxidant-scavenging, and induction of apoptosis. MRP-8 also acts as danger associated molecular pattern (DAMP) molecule and stimulates innate immune cells via binding to pattern recognition receptors such as Toll-like receptor 4 (TLR4) and receptor for advanced glycation end (AGER) products. It also plays an impotent role as oxidant scavengers and has a protective role in preventing exaggerated tissue damage by scavenging oxidants. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. 外形
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
