Quality Control Testing

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected AUTS2 in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected AUTS2 in lysate from C6 and HepG2 cells and Human brain tissue and in a lysate overexpressing Myc/DDX-FLAG tagged full-length human AUTS2.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected AUTS2 in PC12 cells.
Affinity Binding Assay:: A representative lot of this antibody bound AUTS2 peptide with a KD of 9.5 x 10-8 in an affinity binding assay.

Autism susceptibility gene 2 protein (UniProt: Q8WXX7; also known as AUTS2) is encoded by the AUTS2 (also known as KIAA0442) gene (Gene ID: 26053) in human. AUTS2 is a component of a PRC1-like complex that is strongly expressed in brain, skeletal muscle, and kidney. Its expression is also observed in placenta, lung, and leukocytes. Two major forms of human AUTS2 have been described: full-length (1259 aa) and a truncated C-terminal (711 aa), which is produced from an alternative transcription start site in exon 9. Full-length AUTS2 is localized in both the nucleus and cytoplasm, whereas the truncated AUTS2 isoforms are exclusively nuclear. Cytoplasmic AUTS2 is reported to be involved in cell motility and morphogenesis by regulating reorganization of the actin cytoskeleton. AUTS2 is detected in the cell nuclei of glutamatergic neurons in the cerebral cortex during early developmental stages in mouse and as neural development proceeds, it also appears in the cytoplasm of differentiated neurons, including dendrites, axons, and cell bodies. AUTS2 contains several putative nuclear localization signal sequences and two proline-rich domains. Other predicted protein motifs include a PY (Pro-Tyr) motif (PPPY), eight CAG (His) repeats, several protein kinase phosphorylation sites, and putative SH2- and SH3-binding domains. However, the exact role of these motifs has not been analyzed. AUTS2 activates RAC1 via guanine exchange factors (GEFs) that leads to lamellipodia formation in neuroblastoma cell lines. The proline-rich region is important for the interaction with RAC guanine nucleotide exchange factors and the subsequent activation of RAC1, which then promotes lamellipodia formation. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Avoid repeated freeze-thaws.
