一般描述

XTT(3′-[1-[(苯胺基)-羰基]-3,4-四唑]-双(4-甲氧基-6-硝基)苯-磺酸钠)是一种四唑盐,在生物还原时可生成水溶性甲臜。

应用

XTT钠盐已用于评估细胞活性。
XTT可与吩嗪硫酸甲酯(PMS)联合使用,用于筛选人肿瘤细胞系。 四唑染料也被用于真菌细胞损伤研究 、葡萄球菌的抗菌敏感性测试,以及念珠菌生物膜研究。XTT检测法的局限性也有报道。

生化/生理作用

XTT(3′-[1-[(苯胺基)-羰基]-3,4-四唑]-双(4-甲氧基-6-硝基)苯-磺酸钠)受到代谢活性细胞中存在的脱氢酶的作用。这可产生水溶性甲臜,是一种颜色很深的产物。因为产物是可溶的,不会形成甲臜结晶,这一点与MTT相反,因此XTT在细胞增殖比色法检测中更具优势。

原理

在活细胞中,XTT被代谢还原,生成可比色的水溶性甲臜产物。
