黄嘌呤氧化酶(XO),一种多功能的含钼黄素蛋白,是黄嘌呤氧化还原酶(XORs)的形式之一。每个黄嘌呤氧化酶由2个黄素分子(FAD)、2个钼原子和8个铁原子构成。

应用

黄嘌呤氧化酶(微生物)可用于:
  • 测定体外黄嘌呤氧化酶活性
  • 在超氧化物歧化酶检测中生成超氧阴离子
  • 作为比色纸装置的反应混合液组分,酶法检测传统药物中的别嘌呤醇

生化/生理作用

黄嘌呤氧化酶在嘌呤代谢中催化次黄嘌呤羟基化为黄嘌呤,黄嘌呤羟基化为尿酸和氧自由基。这种酶也参与咖啡因代谢。黄嘌呤氧化酶在药物等异源物质的代谢中也起重要作用,例如硫嘌呤类药物,涉及6-巯基嘌呤、别嘌呤醇和尿酸。黄嘌呤氧化酶活性过高会引起高尿酸血症(血中尿酸过多),进一步导致痛风。
还含有蛋白水解活性。
