一般描述

波形蛋白由人类染色体10p13上的单拷贝基因编码。53kDa的波形蛋白属于III类中间纤维(IF)蛋白质家族,由正常间质细胞表达。含466个氨基酸的波形蛋白特征在于高度保守的a-螺旋“棒状” 结构域,该结构域两侧为非α-螺旋氨基- 末端 “头” 和羧基末端 “尾” 结构域。

特异性

在免疫组织化学染色中,抗体将波形蛋白定位于成纤维细胞、内皮细胞、淋巴组织和黑色素细胞中。它也在肉瘤、淋巴瘤和黑色素瘤中染色波形蛋白。

应用

单克隆抗波形蛋白抗体已用于免疫印迹和免疫细胞化学。
小鼠产生的单克隆抗波形蛋白抗体已用于:

生化/生理作用

在前列腺癌、乳腺癌、子宫内膜癌、中枢神经系统(CNS)肿瘤、恶性黑色素瘤和胃肠道肿瘤(包括胰腺癌、结直肠癌和肝癌)细胞中,均观察到波形蛋白表达上调。因此,波形蛋白可以作为开发癌症疗法的靶标。在血管内皮生长因子受体 1 (VEGFR-1) 诱导的蛋白激酶 G (PKG) 1 信号传导通路激活(刺激心肌细胞肥大消退)中,波形蛋白发挥着至关重要的作用。波形蛋白丝参与多种生理过程,包括迁移、细胞形状维持和间充质细胞的机械应力耐受。波形蛋白与 LARP6(La 核糖核蛋白域家族成员 6)相互作用并稳定 I 型胶原 mRNA,可能在组织纤维化的发展中发挥重要作用。波形蛋白在维持晶状体完整性中起着至关重要的作用,因此,波形蛋白基因突变会导致显性白内障和粉状白内障。
波形蛋白是未成熟神经胶质细胞的主要细胞骨架成分。它是一种中间丝蛋白,可作为多种半胱天冬酶的底物。半胱天冬酶裂解该蛋白时,会分解中间丝并促进细胞凋亡。
波形蛋白有助于保持间充质细胞的细胞结构和组织稳定性。它参与肿瘤发生、EMT(上皮间质转化)和癌症的转移扩散,控制人乳头瘤病毒16型假病毒的感染内化。

储存及稳定性

需要连续使用时,可在 2-8°C 下储存一个月。对于更长期的储存,溶液可以冷冻成工作等分试样。不建议重复冷冻和解冻。不建议储存在“无霜”冰箱中。如果长期储存时出现轻微混浊,使用前通过离心澄清。
