一般描述

酪氨酸羟化酶 (TH) 由定位于人染色体 11p15 的基因编码。TH 是四种相同亚基组成的四聚体,特征是含有一个调节、催化和四聚体化结构域。该酶利用酪氨酸、四氢生物蝶呤 (BH4) 和 O2 作为辅底物,并利用 Fe2+ 作为辅因子。

从嗜铬细胞瘤中纯化并经SDS变性的大鼠酪氨酸羟化酶。

应用

兔抗-酪氨酸抗体可用于蛋白质印迹和免疫组化。

生化/生理作用

酪氨酸羟化酶 (TH) 催化儿茶酚胺神经递质生物合成的第一个限速步骤,即 L-酪氨酸转化为 L-多巴的过程。L-苯丙氨酸对 TH 的抑制作用可能在苯丙酮尿症中发挥关键作用,并可阻止去甲肾上腺素合成。TH 活性可通过磷酸化来调节。TH 表达下调与各种神经精神疾病有关,例如精神分裂症和帕金森病 (PD)。

目标描述

酪氨酸羟化酶(TH)可参与到苯丙氨酸向多巴胺的转化。它可催化儿茶酚胺生物合成途径的初始限速步骤。

外形

含有10mM HEPES,pH 7.5、150mM NaCl、100 μg/mL BSA和50%甘油的100 μL溶液。
