General description

FTO (Fat mass-and obesity-associated gene) is the responsible gene for mouse ‘fused toes′ mutation. An association between FTO genotype and type 2 diabetes has been confirmed. The presence of the FTO rs9939609 A-allele was found to be positively correlated with other symptoms of the metabolic syndrome, including higher fasting insulin, glucose, triglycerides, and lower HDL-cholesterol.
The fat mass and obesity-associated (FTO) gene is mapped to human chromosome 16q12.2. The encoded protein is a nuclear protein and belongs to the AlkB-related non-haem iron- and 2-oxoglutarate-dependent dioxygenase family.

Biochem/physiol Actions

FTO (Fat mass-and obesity-associated gene) is the responsible gene for mouse ‘fused toes′ mutation. An association between FTO genotype and type 2 diabetes has been confirmed. The presence of the FTO rs9939609 A-allele was found to be positively correlated with other symptoms of the metabolic syndrome, including higher fasting insulin, glucose, triglycerides, and lower high-density lipoprotein (HDL)-cholesterol. FTO stimulates adipogenesis, tumorigenesis, and drug resistance of cancer cells. It does so by reducing the levels of messenger RNA N6-methyladenosine (m6A) of certain genes post-transcription. The FTO single-nucleotide polymorphisms (SNPs) are associated with elevated body mass and obesity in humans.
