乳铁蛋白基因定位于人染色体3p21.31。
乳铁蛋白是一种存在于外分泌和多形核中性粒细胞次级颗粒中的糖蛋白。它是牛奶中的铁结合蛋白。人乳铁蛋白属于转铁蛋白家族。研究表明,它在几种腺上皮细胞和皮肤表皮角质形成细胞中表达。
应用
来自人乳中的乳铁蛋白已被用作生成标准曲线的标准品,用于在酶联免疫吸附试验(ELISA)中对重组型乳铁蛋白进行定量。它还在十二烷基硫酸钠(SDS)-聚丙烯酰胺凝胶电泳(PAGE)中被用作阳性对照。
生化/生理作用
目前确知乳铁蛋白可调节肠道铁的吸收并参与抵抗细菌的防御。它通过激活核因子-B(NF-B)在先天免疫系统中发挥作用。由于乳铁蛋白干扰脂多糖刺激的 TLR4 信号,该激活通过 Toll 样受体 4 途径发生。它可作为人皮肤角质形成细胞和成纤维细胞中低密度脂蛋白受体相关蛋白 1(LRP1)的配体。它能够刺激各种细胞系中的细胞生长。研究表明,乳铁蛋白也参与伤口愈合。
