General description
大多数 MMPs(基质金属蛋白酶)作为无活性的前体蛋白分泌,被细胞外蛋白酶裂解时被激活。基质金属蛋白酶-9 又称 92kDa IV型胶原酶,92 kDa 明胶酶/明胶酶 B (gelatinase/gelatinase B,GELB),CLG4B,由中性粒细胞、巨噬细胞和编号转化细胞分泌,是结构域结构及其活性调控方面最复杂的家族成员。在结构上,基质金属蛋白酶-9 可分为 5 个不同的结构域:激活后裂解的前结构域、由 3 个连续的纤维连接蛋白 II 类型单位组成的明胶结合结构域、含有锌结合站点的催化结构域、富含脯氨酸的连接区和羧基末端血红素样结构域。

Biochem/physiol Actions
基质金属蛋白酶 (matrix metalloproteinase,MMP) 家族的蛋白参与了正常生理过程中细胞外基质的分解,如胚胎发育、繁殖和组织重塑,以及疾病过程,如关节炎和转移。对恒河猴的研究表明基质金属蛋白酶-9 参与 IL-8(白细胞介素-8)诱导的骨髓造血祖细胞的动员,鼠类研究表明其具有肿瘤相关组织重塑中的作用。血小板反应蛋白、椎间盘蛋白,调节基质金属蛋白酶-2 和-9 的有效水平,它们是 ECM(细胞外基质)重塑的关键效应物。这种酶可降解各种底物,包括明胶、IV 型和 V 型胶原以及弹性蛋白。基质金属蛋白酶-9 参与系统性红斑狼疮、类风湿关节炎、多发性硬化等多种自身免疫性疾病,并被视为潜在的治疗靶点。它还与腰椎间盘突出症和干骺端不典型增生有关。
Physical form
在 pH 值为 7.4 的 PBS 中从 0.22 μm 过滤溶液冻干。通常在冻干前加入5-8%甘露醇或海藻糖作为保护剂。
Preparation Note
打开前离心小瓶。用无菌 PBS (pH 值为 7.4) 复溶至 50μg/ mL 的浓度。切勿涡旋。该溶液可在 2-8°C 下储存长达 1 个月。对于长期存储,建议储存温度为 -20°C。
