RBP4 (retinol binding protein 4) was originally recognized in 2005 as an adipocytokine. It is predominantly produced by hepatocytes and adipose tissues. This gene is localized to human chromosome 10q23–24 and is composed of five exons and six introns. RBP-4 Human Recombinant produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 184 amino acids and having a molecular mass of 21 kDa. The Retinol Binding Protein-4 is purified by proprietary chromatographic techniques.
生化/生理作用
RBP4 (retinol binding protein 4) was initially identified as a cytokine capable of increased liver gluconeogenesis and decreasing insulin sensitivity. In subjects with obesity, impaired glucose tolerance, or type 2 diabetes and in non-obese, non-diabetic subjects with a strong family history of type 2 diabetes, the serum levels of this protein correlate to the degree of insulin resistance. This protein is thought to contribute to the pathogenesis of type 2 diabetes. Studies in obese mice show that fenretinide, a synthetic retinoid, increases the urinary excretion of RBP4, thus, normalizing serum RBP4 levels. This results in improved insulin resistance and glucose intolerance, suggesting that decreasing RBP4 levels might have potential as a therapeutic strategy for type 2 diabetes.
