一般描述
经过测序验证的shRNA慢病毒质粒(pLKO.1-puro)以纯化的质粒DNA形式提供，适于病毒生产和瞬时转染。 DNA在Tris,EDTA (TE)溶液中以每孔40 μl等分提供。 每孔平均每次克隆提供2 μg，范围从400 ng到4 μg。套件附带96孔板，有条形码便于识别。包含RefSeq、基因描述、基因符号、克隆ID、Hairpin序列、位点链接和平板图位置的CD随同基因家族集一起提供。

每个MISSION shRNA克隆基于慢病毒质粒载体pLKO.1-Puro构建，随后转化为大肠杆菌。pLKO.1-Puro载体包含细菌（氨苄西林）和哺乳动物（嘌呤霉素）耐药基因，用于筛选细菌或哺乳动物细胞系插入片段。每个克隆集由平均3-5个构建体组成，这些构建体使用专有算法针对每个目标基因设计。因此，当使用这些克隆时，可以预期每个基因集至少有一个构建体为>70%的敲低效率范围，从而检查基因功能损失对一系列基因敲低效率的影响。每个shRNA构建体都已克隆和序列验证，以确保与目标基因匹配。
