一般描述

免疫亲和性分离高丰度蛋白(HAP)已被证明是克服蛋白质浓度宽动态范围和实现低丰度蛋白(LAP)检测最有效的方法之一。鸡的多克隆IgY(蛋黄免疫球蛋白)抗体具有独特的优越特性,可对复杂的蛋白质混合物进行高特异性分离。IgY12柱可有效去除血浆中的12种HAP。去除HAP后,中等丰度蛋白(MAP)是获得LAP的另一个障碍,而LAP是临床重要标志物,也是诸多生物学研究领域的热点问题。因此,若想进行有效和精确的LAP检测分析,MAP分离将是其面临的一个全新挑战。

为应对这一挑战,我们进一步开发了SuperMix IgY系统进一步将样品中的MAP去除。SuperMix系统中含有特异结合中等丰度蛋白的IgY抗体,总蛋白样品通过IgY12或IgY14柱后的流出组分再次通过SuperMix柱子时,MAP被结合到柱子上,流出部分中主要是低丰度蛋白,从而实现对LAP的高度富集。

应用

Seppro® Supermix, LC5(5mL 柱)适用于将人血清中等丰度蛋白(MAP)从人血清低丰度蛋白(LAP)中分离。Seppro® IgY12 或 IgY14亲和性分离去除特异性人血清高丰度蛋白(HAP)。

  1. How does the storage temperature relate to shipping conditions?

    The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.   2. Which document(s) contains shelf-life or expiration date information for a given product?

    If available for a given product, the recommended re-test date or the expiration date can be found on the Certificate of Analysis.

  6. My protein sample contains SDS.  Is this compatible with Product SEP060, Seppro® Supermix LC5?  

    No. We do not recommend using protein samples containing SDS with Product No. SEP060. SDS is a detergent and would be expected to affect adversely the activity of the conjugated antibody in terms of depletion.

  7. I originally purchased my Seppro SuperMix LC5 column from Genway.  Can I still contact Genway for technical support?

    All inquiries on the Seppro technology are now to be referred to Sigma-Aldrich. Customers should not contact Genway for technical assistance.

  8. My question is not addressed here, how can I contact Technical Service for assistance?
