一般描述

菲律宾菌素复合物,是从菲律宾链霉菌 ( S. filipinensis)培养物中分离得到异构多烯大环内酯类物质的统称,而菲律宾菌素 III 是该复合物的主要部分。[1]

应用

Filipin 已被用在双重染色程序中,作为检测聚丙烯酰胺凝胶中脂蛋白的探针,并固定在硝酸纤维素膜上。它也借助于特定的荧光复合物广泛用于定位和定量未酯化胆固醇。
菲律宾菌素复合物制成溶液可用于 HeLa 细胞染色,进行细胞骨架成像。[2]

生化/生理作用

菲律宾菌素是具有抗真菌功能的抗生素。[3]研究发现,菲律宾复合物可通过与膜固醇、麦角固醇和胆固醇相互作用,改变细胞膜结构。[1]与胆固醇结合后,菲律宾菌素呈荧光偏移。因此,菲律宾菌素复合物可用于诊断尼曼匹克(Niemann-Pick) C型病,也是检测和定量细胞膜中胆固醇以及定量脑中脂质的有用工具。[3]

其他说明

Filipin 复合物溶液对空气和光线非常敏感。接收后分装并储存于-20°C 条件下。避免冻融循环。
根据使用的具体方案,使用不同浓度的 Filipin 复合物。根据所用方案,用适当缓冲液稀释 5 mg/mL 贮备液。
Filipin 与胆固醇的复合物相互作用改变了吸收光谱和荧光光谱,使用 340-380 nm 处的激发和 385-470 nm 处的发射(Filipin 荧光染色光漂白非常迅速,因此应立即分析样品)的荧光显微镜进行观察。
