Replication protein A1 (RPA1) is a constituent of the replication protein A (RPA) heterotrimer. RPA1 gene codes for RPA-70KD, the largest subunit of the RPA. RPA1 gene is located on human chromosome 17p13.3.

Replication protein A1 (RPA1) is essential in homology-directed recombinational repair. It plays a key role in nucleotide excision repair, mismatch repair, and base excision repair. RPA1 participates in DNA replication and recombination events. Decreased RPA1 levels can impair the activation of DNA damage checkpoint. Overexpression of the RPA1 gene affects the stability of the genome and results in 17p13.3 duplication syndrome.
