Biochem/physiol Actions

PCNA可识别DNA聚合酶的酸性、非组蛋白的辅助蛋白，因此，也称为聚合酶δ辅助蛋白。固定持续时间可显著影响PCNA免疫反应性的强度。然而，固定延迟不会影响免疫反应性。酶处理会破坏染色。在丙酮-甲醇或甲醇固定细胞的免疫细胞化学标记中，抗体在整个细胞核（核仁和核质）中显示出颗粒染色。在各种正常和恶性组织的增殖细胞核中，特别是在生发中心，观察到特异性染色。

General description

单克隆抗增殖细胞核抗原（小鼠IgG2a同种型）来源于PC 10杂交瘤，该杂交瘤是由小鼠骨髓瘤细胞与PCNA-蛋白A融合蛋白免疫的BALB / c小鼠脾细胞融合产生的。增殖细胞核抗原（PCNA，36 kDa）也称细胞周期蛋白，是DNA聚合酶的辅助蛋白。该蛋白存在于整个细胞周期中不断循环的细胞的核质中。
增殖细胞核抗原(PCNA)是结构和功能保守的DNA滑动钳(β clamps)家族成员。它们在真核生物中为环状复合物-同源三聚体形式，具有近似的六边对称结构。PCNA单体由两个相似的球状结构域组成，通过域间连接环连接，这是一个长的且可能柔性的环。这些单体以首尾相接的方式成环，由α螺旋的正电荷内表面和β折叠的外表面组成。
